An Ion Selective Electrode measures the potential of a specific ion in solution. This potential is measured against a stable reference electrode. The potential difference depends upon the activity of the specific ion, which is related to its concentration.

Standard solutions of known concentrations are prepared and measured with a pH/mV meter. A graph of concentration vs. mV reading is plotted. The unknown solution is measured, and its mV value is located on the graph to determine the corresponding concentration. Dedicated ion meters can construct this calibration curve automatically.

ISEs are sensitive to some other ions to some extent. The Selectivity Coefficient expresses the ability to distinguish between different ions. If the primary ion is A and interfering ion is B, a coefficient of 0.1 means the electrode is ten times more sensitive to A than to B. If the coefficient is 1, it is equally sensitive to both.
